In order to better cooperate with the prevention and control of epidemic situation in COVID-19 and ensure that clinical laboratories can carry out the nucleic acid detection of infectious pathogens such as COVID-19 in a standardized, safe and effective way, the "Training Course for COVID-19 Nucleic Acid Detection Laboratory Technicians" was successfully held on April 28 -29, 2020, entrusted by Jiangsu Provincial Health and Wellness Committee. The number of medical institutions and independent medical laboratories in the province is 188 1 person, of which 15 13 people participated in online answering questions, and 1422 people passed theoretical training.

This training course invited experts from the province to give a special report on infectious disease detection in COVID-19, covering the basic principle of nucleic acid detection of infectious diseases, the principle and application of main technical platforms, the establishment and management of standardized laboratories, biosafety protection and hospital infection control, technical operation specifications, quality control measures, correct interpretation and reporting of results, etc. Professor Xu Bin, director of Jiangsu Clinical Laboratory Center, gave a report on "Detection Regulations and Strategies in COVID-19", explained the relevant legal documents in novel coronavirus, emphasized that single marker and single detection were preliminary results, and virus markers needed to be verified and confirmed, and introduced the standardized reporting process of COVID-19 detection, especially pointing out that COVID-19 could not carry out mixed sample detection.

Professor Xie Jingxin, secretary of the Institute of Occupational Disease Prevention and Control of Jiangsu Provincial Center for Disease Control and Prevention, gave a report on Biosafety Management of Infectious Disease Nucleic Acid Detection Laboratory, and expounded the biosafety management and existing misunderstandings of the laboratory from the aspects of laboratory identification, physical preventive measures, power sockets, communication equipment, emergency treatment, biosafety cabinets, laboratory pressure, disinfection and sterilization, and waste disposal. Professor Jiang, director of the infection management office of Gulou Hospital affiliated to Nanjing University School of Medicine, gave a report on the prevention and control of nosocomial infection in COVID-19, introduced the epidemiological characteristics of infection, explained the laboratory biosafety measures and precautions in detection, and finally answered the common questions in detection. Professor Wu Guoqiu, Dean of Zhong Da Hospital affiliated to Southeast University, gave a report on "Sample Collection, Processing and Nucleic Acid Extraction before Nucleic Acid Analysis in COVID-19", which was explained from five aspects: inspection application, sample sampling, sample transportation, receiving and preservation, sample pretreatment and RNA extraction and purification methods. He said that choosing a suitable sampling tube and paying attention to the sampling opportunity and location can reduce false negatives, and suggested that pretreatment at 56℃ should be used for 30 minutes to reduce the occupational exposure of laboratory personnel. Professor Zhou Guohua, Director of Pharmacy Department of East Theater General Hospital, gave a report on "Methods, principles, characteristics and clinical application of nucleic acid detection in COVID-19", expounded the principle, development and application of PCR in nucleic acid detection in COVID-19, and introduced the reasons and clinical applications of various new technologies and methods such as Qualcomm quantitative gene detection, third-generation sequencing technology and isothermal amplification. Professor Zhao Jianhua from Jiangsu Clinical Laboratory Center gave a report on the Standardization Construction and Quality Assurance of Clinical Gene Amplification Laboratory, and expounded the standardization construction of the laboratory from four aspects: acceptance process, laboratory setup, equipment and personnel, and then expounded the quality assurance of nucleic acid detection in COVID-19 from the aspects of reagent selection and performance verification, standardization of working procedures, quality control and judgment, and result report.

This training course is a special training course for infectious diseases. Technicians who have passed the theoretical training also need to go to the laboratory that has obtained the "Acceptance Certificate of Clinical Gene Amplification Laboratory (Infectious Disease Related Gene Project)" for experimental training, which will be signed by the teaching laboratory teacher and the designated person in charge, and the experimental records and work certificates will be scanned and sent to jsccl@jsccl.com. Only after being approved by the provincial clinical inspection center can you get the certificate. Those who have obtained certificates can only engage in inspection items related to genetic testing of infectious diseases.
